█ Antibody Epitope Mapping Service


Epitope localization has become one of the key elements in vaccine and drug development. Epitope mapping is the process of identifying the binding site of a protein to its corresponding antigen. FDA and EMEA guidelines recommend molecular analysis of interaction sites between drugs and their regulatory filed targets. In addition, a better understanding of interaction sites can help develop more effective drugs and patent-protected products. Antibody epitope mapping is generally categorized into two types:

① Sexual epitopes: small linear peptide chain sequences of about 10-20 amino acids;

② Elephant epitopes: larger regions formed by protein folding. Since the traditional IgG antigenic determining region has a two-pocket structure, this means that an antibody usually has two types of epitopes.

Most of the therapeutic antibodies (drug antibodies) are structural antibodies, also known as spatial conformation antibodies, for which antigenic epitope mapping is commonly verified by DNA recombination techniques. The common practice is to segment the large protein into a fragment size of 50-100 amino acids, then do binding screening with the antibody, and finally obtain the antigenic sequence by LC-MS/MS.
