Affibody Development Services

TekBiotech is committed to providing customers with high-affinity, high-specificity affibody (i.e., bovine Ultralong CDR H3 affibody) development servies, including the discovery of bovine Ultralong CDR H3 affibodies, in vitro activity detection (BLI/SPR/EC50 detection of affibody binding capacity, competitive ELISA detection of affibody neutralizing activity, flow cytometry detection of affibody binding and blocking functions, etc.), in vivo efficacy evaluation (including detection in animal models), providing robust support for clientssubsequent development of therapeutic and neutralizing small-molecule drugs.

Through years of development, TekBiotech has established a comprehensive affibody development system, enabling it to provide clients with high-quality affibody drug discovery services targeting a wide range of targets, including GPCRs, multi-transmembrane proteins, ion channels, proteins with hiddenfunctional epitopes, and viruses.

Affibody Development Services

The affibody development service is a screening service based on bovine antibody preparation technology. Bovines are immunized with engineered antigens (primarily designed to expose the natural functional domains of the target) to generate relevant antibodies. Specific B cells are isolated via flow cytometry, and the Ultralong CDR H3 affibody gene library is generated through multiple rounds of PCR amplification. The relevant affibody sequences are then obtained through NGS sequencing and analysis. The affinity of the affibodies screened by TekBiotech can reach the μM–nM range.

For specific targets, screening methods may vary. For example, when screening for affibodies targeting transmembrane proteins, it is often necessary to incorporate multiple screening approaches—such as cell-based screening and VLP-based magnetic bead screening—to identify affiodies suitable for downstream applications. Scientists at TekBiotech evaluate each client’s project requirements and design the optimal protocol for antigen modification, as well as affibody discovery and screening. The affibody screening procedure is shown in Figure 1:

Step 1: Animal Immunization

(1) Administer the antigen to healthy adult bovine 4–5 times;

(2) Detection the titer using an ELISA;

(3) If the titer meets the requirements, isolate PBMCs or lymphocytes;

10 weeks

Step 2: FACS Sorting and Sequencing

(1) FACS dual-fluorescence sorting of PBMCs or lymphocytes;

(2) Total RNA extraction and cDNA synthesis;

(3) 3 rounds of PCR amplification for identification and product recovery, followed by NGS sequencing and sequence analysis;

6-8 weeks

Step 3: Detection of the Ultralong CDR H3 Affibody

(1) Synthesize the Ultralong CDR H3 affibody;

(2) Detect binding to antigen-overexpressing cell lines via FACS;

(3) Detect affinity via BLI/SPR;

3-4 weeks
